PiggyBac transposon system for reprogramming somatic cells to induced pluripotent stem (iPS) cells.

Induced pluripotent stem (iPS) cells share most, if not all, of the promise of embryonic stem cells, but without the ethical and political issues. They are also more clinically relevant for many applications and are therefore an excellent choice for research in toxicology, regenerative medicine and other areas.

Recently, a DNA transposon called piggyBac (PB) has demonstrated its stem cell reprogramming ability, producing iPS cells with robust expression of pluripotency markers. The PB system is ideal for stem cell research, demonstrating higher efficiency in iPS cell and custom stem cell line generation compared with other technologies, such as viral vectors and plasmids. A unique advantage of the piggyBac reprogramming system is its ability to be removed seamlessly from the genome of custom iPS cells after reprogramming has been completed, leaving no genetic footprint whatsoever, as if the insertion had never occurred. There is no risk of residual genetic alterations that may promote cancers.
